Method synopsis

Deionized water extraction of sediments following Gregorich and Carter (2007) to measure dissolved nutrients and organic carbon. Designed to minimize physical disruption and reflect carbon and nutrient pools available to microbial and root systems.

Synthesized from method descriptions across 7 papers using this protocol.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Metagenome-assembled genomes for oligotrophic nitrifiers From a mountainous gravelbed floodplain (2025), Environmental Microbiology. Implementations in other papers (listed below) may differ.

1. Dissolved inorganic nitrogen measurement
Aqueous phase concentrations of dissolved inorganic nitrogen (DIN) were measured via water extraction by suspending 1 g of sediment in 10 mL of ultrapure water, shaking at room temperature for 2 h, centrifuging at 4,000 rpm for 10-15 min, then filtering the aqueous phase through a 0.22 μm disc filter (Fisher Scientific). DIN (ammonia and nitrate) concentrations were measured on a SmartChem 200 Discrete Analyzer (WestCo Scientific Instruments, Brookfield, CT) by spectroscopy.
Quantities: 1 g sediment in 10 mL ultrapure water, 0.22 μm filterDuration: 2 h shaking, 10-15 min centrifugationConditions: Room temperature shaking, 4,000 rpm centrifugation
