Method synopsis

Standard butterfly collection using aerial nets across multiple sites and sampling periods. Specimens either preserved whole in ethanol or sampled via wing clips before release.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Patterns of genetic variation and local adaptation of a native herbivore to a lethal invasive plant (2024), Molecular Ecology. Implementations in other papers (listed below) may differ.

1. Butterfly collection and sampling
Adult butterflies were collected using aerial nets at 11 sites across East River Valley. In 2016, 100 female and 235 adult butterflies of both sexes were sampled from 5 locations during June-August. In 2019, additional sampling occurred at 7 locations during June-August, collecting specimens for wing clip and midleg samples.
Quantities: 335 total individuals across 11 sites, distances between sites ranged 1-28kmDuration: June-August sampling periods in 2016 and 2019Conditions: Field collections in East River Valley, Gunnison County, Colorado at elevations ca. 2800 to 3400 m.a.s.l.
   Equipment: aerial nets
2. Sample preservation and storage
Whole-body specimens collected in 2016 were stored in 98% ethanol. In 2019, 2mm wing clips were taken from midlegs, butterflies were marked and released. Wing clips and midleg samples were stored in 98% ethanol after collection.
Quantities: 2mm wing clips from butterfliesDuration: Immediate processing after collectionConditions: Specimens preserved in 98% ethanol
   Equipment: 98% ethanol
