Method synopsis

Standard chloroform fumigation direct extraction technique for measuring soil microbial biomass, combined with soil chemistry analysis and enzyme activity assays.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Experimental warming decouples plant-fungal symbiont interactions and leads to a more conservative ecosystem (2026), PNAS. Implementations in other papers (listed below) may differ.

1. Measure soil properties and microbial biomass
Soil resources including plant-available soil nutrients (phosphate, nitrate, and ammonium), soil organic matter, soil pH, and gravimetric soil moisture of each soil sample were quantified from each of the 10-cm increment samples collected above. Soil microbial biomass was extracted using chloroform fumigation direct extraction technique. Microbial enzymes involved in carbon, nitrogen, and phosphorus cycling were assayed.
Quantities: 10 cm increment samples from 1 m depthDuration: Laboratory analysis following field collectionConditions: Laboratory conditions
