Method synopsis
Near-isogenic lines contrasting at the BCMA1/3 locus were transplanted into 16 field environments across Colorado and Idaho to measure fitness components including survival and herbivore damage. Uses a complete block design with multiple CFR-NIL families to control for unlinked loci effects.
Synthesized from method descriptions across 1 paper using this protocol.
Procedure from a recent peer-reviewed implementation
CFR-NILs were transplanted into 16 field environments across Colorado and Idaho. Plants were randomized within each site using a complete block design with at least eight CFR-NIL families to control for possible effects of unlinked loci outside the BCMA1/3 region. Arrays were arranged in three blocks, flush with the surrounding vegetation.
Quantities: 16 environments, 6,860 total CFR-NILs transplantedDuration: Transplants monitored for 4-8 weeks in 2013 and 2015Conditions: Field conditions across elevational gradient in Rocky Mountains
Equipment: field transplant materials
Measure fitness components in field
We scored survival and herbivore damage weekly for the first two weeks of the growing season and then every other day. After eight weeks in field conditions, we censused for survival and herbivore damage. Herbivore damage was scored as percentage of leaf tissue damaged by insects.
Quantities: Measurements on thousands of individual plants across 16 sitesDuration: Weekly for first 2 weeks, then every other day, final census at 8 weeksConditions: Growing season in Rocky Mountain field sites
