Method synopsis

Fourier Transform Infrared spectroscopy analysis of ground needle samples to quantify functional group concentrations including aromatics, ethers, amides, and carbonyls. Uses attenuated-total-reflection accessory for sample analysis.

Synthesized from method descriptions across 2 papers using this protocol.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Effect of elevation, season and accelerated snowmelt on biogeochemical processes during isolated conifer needle litter decomposition (2021), PeerJ. Implementations in other papers (listed below) may differ.

1. Collect needle litter samples
Collected freshly senesced needle litter from Engelmann spruce and lodgepole pine trees at multiple elevation sites
Quantities: Sufficient material for litterbag deployment across all treatment combinationsDuration: During autumn senescence periodConditions: Field collection from healthy mature trees
Equipment: collection bags, field sampling tools

Typical Equipment

• Nicolet iS50 FTIR spectrometer
• attenuated-total-reflection accessory

Output Measurements

• aromatic peak areas
• ether linkage areas
• amide areas
• carbonyl areas
