Measure nectar production rate
Measure nectar production over 48 hours on one or two flowers per plant. Convert measurements to μL/24 h. Take flower color measurements using reflectance spectrometer comparing red vs green reflectance.
Quantities: 1-2 flowers per plant, 48-hour measurement period converted to 24-hour rateDuration: 48 hours per measurement period during 2015-2016Conditions: On same date for given population to control for environmental variation
Equipment: reflectance spectrometer
Measure leaf traits
Measure specific leaf area (SLA) by dividing leaf area by dry mass. Scan leaves with flatbed scanner, count trichomes at 200% magnification using ImageJ, and divide by leaf area. Measure trichome density on one leaf per plant.
Quantities: One leaf per plant for trichome density, leaf area measurements at 200% magnificationDuration: During vegetative growth periodConditions: On fully expanded mature leaves
Equipment: flatbed scanner, ImageJ freeware
Fit clines to trait data
Standardize trait values between 0 and 1 by subtracting minimum and dividing by difference between maximum and minimum across populations. Fit no-tails cline model Y = a + 1/(1+e^(-(X-c)/w)) where Y is standardized trait value, X is distance, a is intercept, c is centre of cline, and w is width of cline.
Quantities: 12 populations per trait, fitted separately for 1991-92 and 2015-16 dataDuration: Post-processing analysis stepConditions: Using Proc NLIN in SAS version 9.3 with Marquardt iterative method
Equipment: SAS statistical software version 9.3
