Method synopsis

Seedlings were grown under different light quality treatments to measure phenotypic plasticity in response to shade cues, specifically changes in red to far-red light ratios that indicate neighboring vegetation.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Heritable plant phenotypes track light and herbivory levels at fine spatial scales (2018), Oecologia. The protocol was originally introduced by Plant community light efficiency: measuring light response as a function of both photosynthesis irradiance. (2014). Implementations in other papers (listed below) may differ.

1. Conduct greenhouse shade avoidance experiment
Grew seedlings from seeds collected from source sites under two light treatments: natural light or filtered light with a lower ratio of red to far-red (R:FR) light using a blue plastic filter (Lee filter 115) that reduces the ratio of R:FR light available to plants. Measured leaf area and petiole length of the four fully expanded and earliest-developing leaves at a single developmental point after emergence of the seventh and eighth leaves.
Quantities: 37 plants total from source sites, 4 leaves measured per plant from families spanning 5 families with 4-11 separate plantsDuration: Growth period until emergence of 7th-8th leaves, then single-timepoint measurementConditions: Two light treatments: natural light vs. Lee filter 115 (blue plastic filter)
Equipment: Lee filter 115 (blue plastic filter), greenhouse growing facilities

Typical Equipment

• Lee filter 115 (blue plastic filter)
• greenhouse growing facilities

Output Measurements

• leaf area
• petiole length
