Method synopsis

Pre-treatment of bittercress plants with jasmonic acid to simulate herbivore-induced plant defenses, followed by controlled bacterial inoculation to test effects on bacterial growth.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Insect herbivory reshapes a native leaf microbiome (2020), Nature Ecology & Evolution. Implementations in other papers (listed below) may differ.

1. Conduct greenhouse infection experiments
Pre-treat bittercress clones with jasmonic acid (1 mM) or mock solution 3 days prior to bacterial inoculation. Randomize 32 distinct bittercress clones to receive one of 12 Pseudomonas strains (6 P. syringae, 6 P. fluorescens) isolated from field sites. Sample leaf discs 2 days post-infection for bacterial abundance analysis.
Quantities: 32 bittercress clones, 12 Pseudomonas strains tested, plants grown for up to 12 months prior to useDuration: 3-day pre-treatment period, 2-day post-infection samplingConditions: University of Arizona greenhouse conditions, plants originally isolated within 2 km of RMBL in 2012
Equipment: jasmonic acid solution (1 mM, Sigma), mock solution, greenhouse facilities

Typical Equipment

• jasmonic acid solution (1 mM, Sigma)
• mock solution
• greenhouse facilities

Output Measurements

• bacterial abundance after treatment
• plant defense response
