Method synopsis

Factorial experimental design using artificial stream channels to test temperature and nutrient effects on algae growth. Solar heating systems create temperature gradients while drip systems deliver controlled nutrient ratios.

Synthesized from method descriptions across 10 papers using this protocol.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Does the morphology of beaver ponds alter downstream ecosystems? (2011), Hydrobiologia. The protocol was originally introduced by Dissolved iron stimulates uptake of organic phosphorus by Didymosphenia geminata (2010). Implementations in other papers (listed below) may differ.

1. Nutrient limitation experiments
Deployed nutrient diffusing substrates upstream and downstream at 10 sites in 2008. Incubated NDS in streams for 17 days and then frozen in film canisters for chlorophyll a extraction. Eight replicates of four treatments deployed: +Nitrogen (N), +Phosphorus (P), +Nitrogen and Phosphorus (N + P), and Control (C). Used KaNO3 for nitrogen additions and KH2PO4 for phosphorus additions. Added nutrients to agar at 0.5 M concentration.
Quantities: 10 sites, 8 replicates per treatment, 4 treatments, 17-day incubationDuration: 17-day incubation period in summer 2008Conditions: In-stream deployment during summer 2008
Equipment: nutrient diffusing substrates, film canisters, KaNO3, KH2PO4, agar
