Method synopsis

Method for collecting fish skin mucus by gentle rubbing in plastic bags followed by freeze-drying and chemical characterization of mucus components including glycosaminoglycans, hexosamine, and uronic acid.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Mayflies avoid sweets: fish skin mucus amino sugars stimulate predator avoidance behaviour of Baetis larvae (2019), Animal Behaviour. Implementations in other papers (listed below) may differ.

1. Collect fish mucus samples
Collected mucus from 10 hatchery-reared Atlantic salmon (mean fork length 16.58 ± 0.3 cm) by gently dabbing fish mouth 10 times with cotton to eliminate excess water, then placing in plastic bag with fresh mucus and gently rubbing each fish for 20 s within the bag, removing fish, and storing bags containing mucus on ice for transport to laboratory.
Quantities: 10 Atlantic salmon, mean fork length 16.58 ± 0.3 cmDuration: 20 seconds rubbing per fishConditions: Fresh mucus stored on ice during transport
 Equipment: cotton, plastic bags
2. Process and characterize mucus
Removed fresh mucus from each bag with micropipette, placed in preweighed vial to estimate mucus amount, lyophilized fresh mucus by freeze-drying each mucus sample in speed-vacuum for 5 h at maximum speed, calculated mucus dry weight and water content, then extracted dry weight lost by lyophilization process.
Quantities: Mucus samples processed individually, 5 hour lyophilizationDuration: 5 hours lyophilizationConditions: Speed-vacuum freeze-drying at maximum speed
Equipment: micropipette, preweighed vials, speed-vacuum freeze dryer
