Method synopsis

Standard protocol for obtaining laboratory-reared larvae from field-collected adult female butterflies through controlled oviposition and larval rearing to desired developmental stage.

Synthesized from method descriptions across 1 paper using this protocol.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Geographic differences and microevolutionary changes in thermal sensitivity of butterfly larvae in response to climate (2014), Functional Ecology. Implementations in other papers (listed below) may differ.

1. Collect adult female butterflies
Adult female butterflies were collected from each site and shipped overnight to laboratory at University of North Carolina at Chapel Hill
Quantities: Sacramento Valley CA: 90 larvae in 296 feeding trials; Chapel Hill NC: 92 larvae in 235 trials; Montrose Valley CO: 168 larvae in 401 trials; Gunnison CO: 134 larvae in 334 trialsDuration: Not specifiedConditions: Field collection during appropriate season for each site
   Equipment: collection materials for butterflies
2. Obtain eggs through oviposition
Female butterflies were kept in cages at greenhouse conditions (26°C) under natural light. Females were fed 10% honey water solution by moistened sponge changed daily and allowed to oviposit on potted Vicia villosa in greenhouse
Quantities: Eggs were reared and fifth instar larvae selectedDuration: Until oviposition occurredConditions: 26°C greenhouse conditions under natural light
Equipment: cages, potted Vicia villosa, honey water solution, moistened sponge
3. Rear larvae to fifth instar
Eggs were reared using standard rearing protocol maintained at 25°C in 14L:10D photoperiod where larvae were given fresh V. villosa leaves
   Reared to fifth instar stage

Typical Equipment
