Method synopsis

Production of F2 hybrid generations with alternate cytoplasmic backgrounds through controlled hand-pollination crosses. Allows testing of cytoplasmic effects on physiological traits.

Synthesized from method descriptions across 2 papers using this protocol.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Leaf physiology reflects environmental differences and cytoplasmic background in Ipomopsis hybrids (2007), American Journal of Botany. Implementations in other papers (listed below) may differ.

1. Produce experimental F2 hybrids
Generated reciprocal F2 hybrids through hand-pollination crosses. F1 hybrids with alternate cytoplasmic backgrounds maintained in pots in RMBL garden, then reciprocally hand-pollinated to produce F2 generation with known cytoplasmic backgrounds.
Quantities: F2T (I. tenuifuba cytoplasm) N=100, F2A (I. aggregata cytoplasm) N=233, total 2630 seeds plantedDuration: Multi-generational crossing program over multiple growing seasonsConditions: Common garden conditions at Rocky Mountain Biological Laboratory
Equipment: hand-pollination tools, greenhouse pots, WeatherPort portable mesh greenhouses

Typical Equipment

• hand-pollination tools
• greenhouse pots
• WeatherPort portable mesh greenhouses
