Method synopsis

Cellulose acetate electrophoresis to analyze genetic variation at enzyme loci in mayfly populations, involving initial enzyme screening, locus selection, and final allele frequency determination across multiple populations.

Synthesized from method descriptions across 2 papers using this protocol.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Are populations of mayflies living in adjacent fish and fishless streams genetically differentiated? (2005), Freshwater Biology. Implementations in other papers (listed below) may differ.

1. Allozyme electrophoresis screening
Used cellulose acetate electrophoresis on Helena Laboratories Titan III plates with stains modified from Richardson, Baverstock & Adams (1986). Initially screened 25 enzymes for variation, identified 6 loci that were both variable and could be reliably interpreted.
Quantities: 25 enzymes initially screened, 6 loci selected for analysisDuration: Not specifiedConditions: Laboratory conditions
Equipment: Helena Laboratories Titan III plates, cellulose acetate electrophoresis system
2. Final allozyme analysis
Determined allele frequencies at six different loci for both species at each site: Glucose phosphate isomerase (PGI, IECC no.5.3.1.9), phosphoglucomutase (PGM, IECC no.2.7.5.1), peptidase PEPB and PEPC (IECC no. 3.4.11), and amino aspartate transferase (AAT, IECC no. 2.6.1.1) which had two variable loci.
Quantities: 6 loci analyzed across all samplesDuration: Not specifiedConditions: Laboratory conditions
   Equipment: electrophoresis equipment, enzyme-specific stains
