Method synopsis

Plant biomass consumption by herbivores is quantified using before-and-after digital photographs with standardized reference objects. Image analysis software calculates the difference in plant material between photos to determine consumption rates.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Pre- and post-ingestive defenses affect larval feeding on a lethal invasive host plant (2019), Entomologia Experimentalis et Applicata. Implementations in other papers (listed below) may differ.

1. Laboratory leaf preparation
Newly hatched larvae placed individually on entire excised leaves in 45-mm-diameter Petri dishes lined with moist filter paper. Used leaves with no visible signs of previous abiotic or biotic damage. Each leaf photographed from fixed distance alongside red calibration square before and after 6-h trial using Leica S6D Greenough stereomicroscope at 2, 4, and 6 h intervals
Quantities: 237 larvae total across trials, 45-mm-diameter Petri dishes, individual larvae per dishDuration: 6 hours total trial time with observations at 2, 4, and 6 hConditions: Growth chamber conditions, moist filter paper
Equipment: 45-mm-diameter Petri dishes, moist filter paper, Leica S6D Greenough stereomicroscope, red calibration square
