Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — High-discharge disturbance does not alter the seasonal trajectory of nutrient uptake in a montane stream (2021), Hydrobiologia. Implementations in other papers (listed below) may differ.

1. Conduct nutrient uptake tracer experiments
Inject rhodamine water tracer and either NH4Cl or Na2HPO4 simultaneously at upstream locations, monitor breakthrough curve at downstream endpoint using handheld fluorometer, collect 30-40 water samples during each BTC for nutrient analysis
Quantities: Six uptake measurements per summer for each nutrient, tracer concentrations raised 10-20x above ambientDuration: Each tracer experiment conducted over ~40 width measurements periodConditions: Conducted from June through September during both summers
Equipment: Rhodamine WT, NH4Cl, Na2HPO4, Databank handheld fluorometer, Turner Designs syringes with inline filter holders, Gelman AE glass fiber filters
2. Analyze water samples for nutrient concentrations
Analyze NH4+-N using standard fluorometric method, pre-concentrate SRP samples 70-fold using Sargasso Sea water magnesium hydroxide co-precipitation (MAGIC-P) method, analyze using conventional colorimetry on Thermo Scientific GENESYS 10S UV-VIS Spectrophotometer
Quantities: 30-40 water samples collected per breakthrough curve experimentDuration: Analysis conducted immediately or refrigerated until following dayConditions: Samples kept on ice until lab analysis
Equipment: Turner Designs Trilogy Fluorometer Model #7200, Thermo Scientific GENESYS 10S UV-VIS Spectrophotometer, MAGIC-P pre-concentration materials
