Method synopsis

Wood samples are sectioned into sub-annual increments and cellulose is extracted for oxygen and carbon isotope analysis to assess seasonal water sources and water use efficiency in trees.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Depth of nutrient uptake by deep-rooted plants is regulated by water availability (2026), PNAS. The protocol was originally introduced by Observed forest sensitivity to climate implies large changes in 21st century North American forest growth (2016), Ecology Letters. Implementations in other papers (listed below) may differ.

1. Collect and analyze sagebrush growth rings
One sagebrush plant from each of two study locations was sectioned for growth rings. Growth ring samples were mounted, sanded, and measured for ring width using a sliding scale micrometer. Ring samples were dissolved in HNO3 and analyzed for 87Sr/86Sr ratios by ICP-MS.
Quantities: 1 plant per site, multiple growth rings per plant spanning ~30 yearsDuration: Single collection, laboratory analysis over multiple daysConditions: Laboratory preparation and analysis
Equipment: sliding scale micrometer, mounting materials, sanding equipment, ICP-MS

Typical Equipment
