Method synopsis

Plants were subjected to progressive water stress by withholding irrigation to create a gradient of soil moisture conditions. This experimental design allowed examination of plant responses across a range of drought severity levels.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — Phenotypic plasticity of floral volatiles in response to increasing drought stress (2019), American Journal of Botany. The protocol was originally introduced by Photosynthetic and growth responses of reciprocal hybrids to variation in water and nitrogen availability (2010), American Journal of Botany. Implementations in other papers (listed below) may differ.

1. Obtain and establish study plants
Flowering individuals of I. aggregata were obtained from sub-alpine meadows 1 km south of the Rocky Mountain Biological Laboratory, Gunnison County, CO, USA. F₂ hybrid plants originating from a cross between I. aggregata subsp. aggregata and I. tenuituba were also included. Plants were moved into an outdoor WeatherPort greenhouse and watered ad lib for 2 weeks prior to use in experiments.
Quantities: 8 plants in 2015, 10 plants in 2017, total of 18 plants across both yearsDuration: 2 weeks recovery period after transplantConditions: Outdoor WeatherPort greenhouse conditions
Equipment: WeatherPort greenhouse, 6 inch plastic pots
2. Implement progressive drought treatment
Plants were allowed to dry out progressively after withholding irrigation. The experiment generated a range of soil moistures typical of those experienced by local natural populations, from 2.7-26% volumetric water content (vwc). Both plant-level gas exchange and floral volatiles were measured at four time points during the drought.
Quantities: Range of 2.7-26% volumetric water content, 4 measurement time points per plantDuration: Progressive dry-down over approximately 13 days in each experimentConditions: Outdoor conditions, experiments conducted in 2015 and 2017
