Method synopsis

DNA extraction from hair samples followed by microsatellite genotyping to determine parentage relationships and construct molecular pedigrees for quantitative genetic analysis.

Synthesized from method descriptions across 19 papers using this protocol.

Procedure from a recent peer-reviewed implementation

Steps below were extracted from the most recent peer-reviewed implementation of this protocol in the corpus — The heritability of fear: decomposing sources of variation in marmot flight initiation distance (2025), Animal Behaviour. Implementations in other papers (listed below) may differ.

1. DNA extraction and molecular genealogy
Hair samples were stored in envelopes and kept at -20°C. DNA was extracted using the QIAmp Mini Kit by Qiagen Inc. and amplified via polymerase chain reaction (PCR). Microsatellite genotyping was performed at 12 loci to match parents to offspring and analyse allele frequencies using GENEMAPPER. Parentage was assigned using CERVUS 3.0, resulting in samples having 95% confidence in parental assignment.
Quantities: 12 microsatellite loci used, 95% confidence threshold for parental assignmentDuration: DNA samples collected from 2001 onwardsConditions: Hair samples stored at -20°C
Equipment: QIAmp Mini Kit by Qiagen Inc, GENEMAPPER software, CERVUS 3.0 software
